Effective Design of Multifunctional Peptides by Combining Compatible Functions
نویسندگان
چکیده
Multifunctionality is a common trait of many natural proteins and peptides, yet the rules to generate such multifunctionality remain unclear. We propose that the rules defining some protein/peptide functions are compatible. To explore this hypothesis, we trained a computational method to predict cell-penetrating peptides at the sequence level and learned that antimicrobial peptides and DNA-binding proteins are compatible with the rules of our predictor. Based on this finding, we expected that designing peptides for CPP activity may render AMP and DNA-binding activities. To test this prediction, we designed peptides that embedded two independent functional domains (nuclear localization and yeast pheromone activity), linked by optimizing their composition to fit the rules characterizing cell-penetrating peptides. These peptides presented effective cell penetration, DNA-binding, pheromone and antimicrobial activities, thus confirming the effectiveness of our computational approach to design multifunctional peptides with potential therapeutic uses. Milk proteins provide a natural source of bioactive peptides with potential health benefits and applications in the food industry. The release of these peptides from milk proteins is achieved either by hydrolysis using digestive proteases or by lactic acid bacteria fermentation. Peptides, particularly those derived from milk proteins, can exert a wide range of nutritional, functional and biolog...
متن کاملFluorescent, Magnetic Multifunctional Carbon Dots for Selective Separation, Identification, and Eradication of Drug-Resistant Superbugs
The emergence of drug-resistant superbugs remains a major burden to society. As the mortality rate caused by sepsis due to superbugs is more than 40%, accurate identification of blood infections during the early stage will have a huge significance in the clinical setting. Here, we report the synthesis of red/blue fluorescent carbon dot (CD)-attached magnetic nanoparticle-based multicolor multif...
متن کاملEffective Design of a 3×4 Two Dimensional Distributed Amplifier Based on Gate Line Considerations
In this paper two dimensional wave propagation is used for power combining in drain nodes of a distributed amplifier (DA). The proposed two dimensional DA uses an electrical funnel to add the currents of drain nodes. The proposed structure is modified due to gate lines considerations. Total gain improvement is achieved by engineering the characteristic impedance of gate lines and also make appr...
